16.10 Keynote “New Insights on Microclots”
Amyloid fibrin microclots (AFMs) in human blood are increasingly recognized as significant indicators of health status, particularly in conditions such as post-acute COVID-19 syndrome (PACS), post-COVID-19 vaccination syndrome (PCVS), myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS), and in patients dealing with chronic inflammation. These microclots, which form due to vascular damage or exposure to inflammatory molecules, are commonly linked to various inflammatory diseases, with affected individuals showing increased numbers and sizes of AFMs. At INUS Laboratories, we have developed a novel method to detect and quantify AFMs using imaging flow cytometry. This protocol, optimized for platelet-poor plasma analysis, enables reliable detection of AFMs in blood samples processed within 24 hours of collection. To facilitate accurate analysis, a data framework was developed to define the size distribution of AFMs. Our method shows great potential for disease diagnosis and therapy monitoring. Despite logistical challenges, the ongoing development of this tool represents a significant advance in understanding and managing inflammation-related conditions.
